I've been watching Roseanne over the past few months. I'm in the last few episodes of the series now and the one thing that has remained consistent for me is my love of Mark. The guy is such a lovable, funny character. A bit of a doofus, but the kind of guy I'd want to be friends with, a guy with heart. Makes me sad that Glenn's life ended the way it did.
Thinking about that made me wonder who all you guys enjoy in supporting roles on TV and why.

John Locke from LOST. Easily one of the most compelling characters in tv history.

Kramer. You could call him a main character but you could also say Jerry Seinfeld is the star and George, Kramer and Elaine are supporting main characters.

If I have to narrow it down to one I'll say Henry Deacon from Eureka. The character went through a wonderful arc throughout the series and Joe Morton did a great job in the role.
Can't let it go without at least mentioning Radar from MASH too.

Anya, Giles and Spike - Buffy the Vampire Slayer.
Wesley, Doyle, and Lorne - Angel.
Bobby, Jody Mills, and Crowley - Supernatural.
Walter Skinner and The Lone Gunmen - X-Files.
I could probably go on, but these are some of the main ones that immediately come to mind.

Coach, Norm and Cliff from Cheers. They brought the funny and the heart.
Fargo from Eureka. He always stole the show with his awkwardness and his love for Jo.
TLG from X-Files. No explanation needed. Frohike and Dana should have happened.
Taco from the League. No one was funnier than him, ever.

Ben Linus from LOST - LOST is my fave show of all the times and I could mention a dozen or so supporting characters that are amazing in their own right. Ben Linus tho is one that I struggled with at first but by the end he wound up being one of my favourites. My god, the performance from Michael Emerson throughout the show is one of the best. Ben is a fascinating character to watch, unpredictable and emotionally engaging as a figurehead who just doesn't know how he fits into the world unravelling around him. Has some of the most powerful scenes in the shows entire run.
