On occasion. Anything with pets or parents or the elderly or lost love or goodbyes can get to me. Nothing having to do with children ever does though. The worst offender is probably the Smallville episodes Reckoning and Vengeance. Jonathan Kent Dies at the end of Reckoning, but the end of Vengeance has Clark and Martha watching an old family video. It ends with Jonathan saying "Goodbye son... goodbye" and riding off on a tractor as Clark and Martha cry. I am tearing up just typing this. The fact that my dad died a few years ago doesn't help. Requiem from Who's the Boss, which I saw for the first time about a year ago, for the same reason.
The Finale of Frasier also did it when he was saying goodbye to everyone...and the Tenth Doctor's farewell tour prior to his regeneration, as cheesy as it was.
As far as a single manly tear or misty goes, the part of the Day of the Doctor when Tom Baker insinuated that the Doctor one day retires, becomes the Curator and can revisit his old faces.
I expect the finale of HIMYM to do it too, regardless of whether or not the mother is dead. It was pretty close during Vesuvius, and has already happened a few times this season. Ted finally letting go of Robin did it, as did the mother's rendition of La Vie En Rose. Something about it.
Filmwise, Rocky III with Mick's death, Rocky IV with Apollo's, and......yeah, the Notebook.

And while I'm on the subject of it, the episode was a bit corny but the How to Save a Life moment from Grey's Anatomy always got me.
