Are we ruling out characters that later went on to be billed as stars?
Either way, Faith from Buffy and Angel immediately springs to mind.

Yeah, definitely Faith in Buffy and Angel. She's my favorite slayer by far in the entire Buffy U. I would say Spike as well. I mean he became a main stay in later seasons, of both shows, but he was always great when he showed up early on. Those are two of my favorite TV characters ever, period.
Obviously Odenkirk as Saul Goodman in Breaking Bad.
I really liked Craig Owens in Doctor Who. Thought he was a quirky and fun character. I think we only got 2 episodes with him, but I thought they were both great. I'd say Saffron in Firefly falls into this same category for me where we didn't see her more than twice but she was a great character.
Paul Rudd on Friends and Kyle MacLachlan on HIMYM. Definitely Rafi on the League.

Definitely Rafi on the League. Oh yeah, Mike (Rudd) on Friends is a great one. I'd also put David (Hank Azaria) up there too. Friends had a LOT of great ones though. Greg Kinnear, Tom Selleck, Aisha Tyler and the guy that played Eddie were all great too. Spike is a great character, but a lot of the time it just seemed as if he's there because he is a popular character. Like they didn't really know what to do with him a lot of the time. I wish Faith was used more, she and Doyle are easily my favorite characters from Buffy and Angel respectively (hence the Avatar/Sig).

As my avatar and signature will probably help you to deduce, my list will consist entirely of Doctor Who characters. River Song (played by Alex Kingston) - Such a terrific character. Whether she's flirting with The Doctor, or killing a whole roomful of the Silence by herself, or giving nothing away about the future, she is played brilliantly by Alex Kingston. Craig Owens (played by James Corden) - Considering that I was already a fan of his work before he was in Doctor Who, I was glad to hear that James Corden would be in Doctor Who. He then went on to appear again. Since James Corden is a brilliant comedy actor (he can also do serious very well too), I'd love to see him reappear in Doctor Who. Sabalom Glitz (played by Tony Selby) - Even though I've only seen 2 out of the 3 episodes he has appeared in, I love the character. Even though he's an entirely selfish psychopath, he acts in such a way that he is funny and charming.
